Preclinical ADC development can be complicated by differences in antigen expression, antibody binding, and tumor biology that are difficult to capture in conventional models.
In this AACR 2026 video poster presentation, see how the RASTRUM Allegro platform was used to generate high-throughput, patient-derived 3D colorectal cancer tumoroid models for testing a CEACAM5-targeted antibody-drug conjugate across two CRC subtypes.
The bioprinted models maintained distinct molecular and phenotypic features and enabled comparison of ADC response with standard-of-care agents.
Despite comparable CEACAM5 expression, the two models showed markedly different ADC responses. The responsive HuCo021320 model showed potent activity with labetuzumab govitecan, while HuCo031721 showed minimal response, consistent with its apparent intrinsic resistance to the SN-38 payload.
